  1. Stimulation Phase: Controlled ovarian stimulation is performed using medications to promote the development of multiple follicles.

  2. Triggering Ovulation: Human Chorionic Gonadotropin (hCG) or GnRH agonist is administered to mature oocytes and trigger ovulation.

  3. Retrieval Procedure: Oocytes are collected using a trans vaginal ultrasound-guided aspiration technique. The procedure is minimally invasive and performed under sedation.


Handling Oocytes in the Lab


  1. Collection and Identification: Follicular fluid is collected and examined under a microscope to identify oocytes.

  2. Washing and Grading: Oocytes are washed in culture media to remove debris and are graded based on maturity (e.g., germinal vesicle, metaphase I, metaphase II).

  3. Culture: Mature oocytes (MII) are placed in specialized culture media and incubated at optimal conditions.


Protocols for Oocyte Handling


  1. Temperature Control: Maintain oocytes at 37°C to avoid thermal stress.

  2. Minimized Light Exposure: Protect oocytes from photo damage by limiting light exposure during handling.

  3. Sterile Techniques: Use sterile tools and media to prevent contamination.

  4. Documentation: Record all procedures, observations, and outcomes for quality control.
